The image, Figure 6.14 from The Cell: A Molecular Approach. 2nd edition, depicts a gene going through In vitro splicing. The gene, containing an intron is cloned downstream of a promoter(P) recognized by a phage RNA polymerase. The plasmid digested with a restriction enzyme, cuts the 3’ end of the inserted gene, resulting in a linear DNA molecule. Then, the DNA is transcribed in vitro with the phage polymerase to produce pre-mRNA. The splicing reactions can then be studied in vitro with the addition of the pre-mRNA to nuclear extracts of mammalian cells.
